Ⅰ型補體受體抗體,Anti-CD35抗體產品介紹：This gene is a member of the receptors of complement activation (RCA) family and is located in the 'cluster RCA' region of chromosome 1. The gene encodes a monomeric single-pass type I membrane glycoprotein found on erythrocytes, leukocytes, glomerular podocytes, and splenic follicular dendritic cells. The Knops blood group system is a system of antigens located on this protein. The protein mediates cellular binding to particles and immune complexes that have activated complement. Decreases in expression of this protein and/or mutations in its gene have been associated with gallbladder carcinomas, mesangiocapillary glomerulonephritis, systemic lupus erythematosus and sarcoidosis. Mutations in this gene have also been associated with a reduction in Plasmodium falciparum rosetting, conferring protection against severe malaria. Alternate allele-specific splice variants, encoding different isoforms, have been characterized. Additional allele specific isoforms, including a secreted form, have been described but have not been fully characterized.

抗原修復的幾種常用方法（供參考）

1、 微波爐加熱：

在微波爐里加熱0.01*緩沖液（pH6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復1-2次。根據玻片情況可參考采用微波加熱“3-5-3法”3分鐘溫火沸騰后靜止5分鐘，再溫火沸騰3分鐘即可。

適用的抗原：來源于人、大鼠、小鼠的組織標本；來源于其他動物種屬的組織標本。

2、 沸熱修復：

電爐或水浴鍋加熱0.01M*緩沖液（pH6.0）至95℃左右，放入組織芯片加熱10-15分鐘。

3、 高壓熱修復：

在沸水中加入EDTA（pH8.0）或0.01m*緩沖溶液（pH6.0）.蓋上不銹鋼鍋蓋，但不能鎖定。將玻片置于金屬染色架上，緩慢加壓，使玻片在緩沖液中浸泡五分鐘，然后將蓋子鎖定，小閥門將會升起來。10分鐘后除去熱源，置入涼水中，當小閥門沉下去后打開蓋子。此方法適用于較難檢測或核抗原的抗原修復。（骨及軟骨組織的標本選擇較溫和的微波加熱修復） 。

4、 酶消化方法：

常用0.1%*和0.4%*液。*使用前預熱37℃，消化時間約為5-30分鐘。

適用于被固定遮蔽的抗原：包括 Collagen.GFAP. Complement.Cytokeratin.c-erB-2.LCA.LN.等。

抗體修復方式:

修復液：0.01M 枸櫞酸（pH6.0）或者0.05M EDTA（pH8.0）

抗體修復方法：

方法1：沸水浴修復，將盛有修復液和玻片的燒杯置于沸水浴環境，保持外部沸騰狀態15min，自然冷卻至室溫；

方法2：微波修復，將盛有修復液和玻片的燒杯置于微波爐中，高火5min，停火3min，中火5min，自然冷卻至室溫。
