Anti-CK20抗體說明書；細胞角蛋白20抗體產品介紹：Cytokeratin 20 is a type I keratin which is primarily expressed in gastric and intestinal epithelium, urothelium, and Merkel-cells. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Their genes are clustered in a region of chromosome 17q12-q21. Cytokeratin 20 is a major cellular protein of mature enterocytes and goblet cells and is specifically expressed in the gastric and intestinal mucosa. It is also expressed in adenocarcinomas of the colon, stomach, pancreas and the bile system and is present in mucinous ovarian tumors, transitional-cell and Merkel-cell carcinomas. Notably, the squamous cell carcinomas and adenocarcinomas of the breast, lung, and endometrium, non-mucinous tumors of the ovary, and small cell carcinomas lack cytokeratin 20.

福爾馬林固定的組織有可能破壞了原來組織的抗原結構，蛋白多肽發生交聯，導致部分抗原表位封閉，結合位點減少，所以需要抗原修復，以暴露原來的抗原表位，達到充分顯露抗原，以不出現明顯脫片現象為佳。

抗原修復的幾種常用方法（供參考）

1、 微波爐加熱：

在微波爐里加熱0.01*緩沖液（pH6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復1-2次。根據玻片情況可參考采用微波加熱“3-5-3法”3分鐘溫火沸騰后靜止5分鐘，再溫火沸騰3分鐘即可。

適用的抗原：來源于人、大鼠、小鼠的組織標本；來源于其他動物種屬的組織標本。

2、 沸熱修復：

電爐或水浴鍋加熱0.01M*緩沖液（pH6.0）至95℃左右，放入組織芯片加熱10-15分鐘。

3、 高壓熱修復：

在沸水中加入EDTA（pH8.0）或0.01m*緩沖溶液（pH6.0）.蓋上不銹鋼鍋蓋，但不能鎖定。將玻片置于金屬染色架上，緩慢加壓，使玻片在緩沖液中浸泡五分鐘，然后將蓋子鎖定，小閥門將會升起來。10分鐘后除去熱源，置入涼水中，當小閥門沉下去后打開蓋子。此方法適用于較難檢測或核抗原的抗原修復。（骨及軟骨組織的標本選擇較溫和的微波加熱修復） 。

4、 酶消化方法：

常用0.1%*和0.4%*液。*使用前預熱37℃，消化時間約為5-30分鐘。

適用于被固定遮蔽的抗原：包括 Collagen.GFAP. Complement.Cytokeratin.c-erB-2.LCA.LN.等。

修復液：0.01M 枸櫞酸（pH6.0）或者0.05M EDTA（pH8.0）

抗體修復方法：

方法1：沸水浴修復，將盛有修復液和玻片的燒杯置于沸水浴環境，保持外部沸騰狀態15min，自然冷卻至室溫；

方法2：微波修復，將盛有修復液和玻片的燒杯置于微波爐中，高火5min，停火3min，中火5min，自然冷卻至室溫。
